Coliforms 100 ReadyCult® pkg of 20 tests
Coliforms 100 is a chromogenic medium that provides easy-to-read, definitive positive results. It has a high nutritional quality of the peptones along with incorporated phosphate buffer that guarantees rapid growth of coliforms, while lauryl sulfate largely inhibits the accompanying flora (especially the Gram-positive). The chromogenic substrates X-GAL (cleaved by coliforms) and the fluorogenic substrate MUG (highly specific for E. coli) allow for the simultaneous detection of total coliforms and E. coli. The presence of total coliforms is indicated by a blue-green color of the broth and E. coli by a blue fluorescence under UV-light. The optional 30-second indole reaction helps to accurately confirm the presence of E. coli. ReadyCult® tests are convenient, rapid and economical.
Application
Coliforms 100 is a rapid E. coli/Coliform test for drinking water. It is a selective enrichment broth used for the simultaneous presence / absence detection of total coliforms and E. coli within the bacteriological water examination.

Incubation: 24 hrs.; 35 °C; aerobic.
- Sample Addition: Pour or collect 100 ml of the water sample into a sterile, transparent, chemically clean sampling bottle.
- Inoculation: Open one ReadyCult® Coliforms 100 snap pack carefully and empty the dry broth medium into the 100 ml water sample.
- Mixing: Close the bottle securely and swirl or shake gently until the medium is completely dissolved.
- Incubation: Place the bottle in an incubator set to 35°C–37°C for 18 to 24 hours.
- Negative Result: The yellowish color of the medium remains unchanged, or turbidity appears without a color change.
- Total Coliforms (Positive): The color of the liquid changes from yellowish to blue-green.
- Escherichia coli (E. coli): Illuminate the blue-green sample with a UV light (366 nm). A light-blue fluorescence indicates the presence of E. coli.
- Optional Indole Confirmation: Add Kovacs’ reagent to the tube for a quick 30-second indole test to further confirm E. coli if fluorescence results are unclear or require verification.
